AmplifyRP XRT for Flavescence dorée (FD) is a rapid DNA amplification and detection platform designed for testing grapevines for Candidatus Phytoplasma vitis Flavescence dorée (16SrV-D). This kit includes lyophilized reaction pellets containing the necessary reagents to amplify FD DNA and an exogenous DNA control at a single operating temperature (42 °C).
AmplifyRP XRT is a real-time isothermal DNA amplification and detection system that rapidly amplifies small portions of DNA and offers unrivaled detection capabilities in an easy-to-use testing format. It offers comparable sensitivity and specificity to published PCR methods while eliminating laborious and costly nucleic acid extractions.
The test can be performed virtually anywhere using the battery operated AmpliFire fluorometer. Assay parameters are loaded via barcode and results are automatically displayed as ( + ) or ( - ).
Prior molecular diagnostic experience is not required to perform AmplifyRP XRT tests.

Limit of Detection (LOD): 100 genomic copies per reaction
Specificity:
- Inclusive: Detects all Candidatus Phytoplasma vitis Flavescence dorée (16SrV-D) isolates, including FD-D, FD-C, FD-CS, FD-PS, FD-CCI, FD-92, FD-2000, ALY and FD-70.
- Exclusive:
- Does not detect: Phytoplasma solani.
- Predicted not to detect: Palatinate Grapevine Yellows (PGY-A, PGY-B, and PGY-C), Hemp dogbane phytoplasma (HD-1), Phytoplasma trifoli, Phytoplasma australiense, Phytoplasma pruni, Phytoplasma asteris, Phytoplasma ulmi, Phytoplasma ziziphi, Phytoplasma balanitae, and Phytoplasma rubi.
Total Assay Time: Less than 30 minutes when used with the AmpliFire as a real-time assay.
